It depends if it's sub lingual or not. The former takes effect almost instantly.
Anyway, if you take 100mg to 200mg of B-6 right before you go to bed, it should make a huge difference. Don't take it every night, so that your body won't start getting a tolerance to it. Also, you should never take high dosages of B-6 for an extended period of time, as it will cause permanent nerve damage. If you take 100mg to 200mg every day, I'm pretty sure you're still safe, but your body would start getting the tolerance, so try to space it out.
